Abstract

Asynchronous molting, cannibalism, and high juvenile mortality limit the production of the redclaw crayfish Cherax quadricarinatus. This study aimed to characterize steroid-related compounds in a methanolic extract of the green mussel Perna viridis and to evaluate the effectiveness of the extract, applied by immersion, in the diet, or both, on molting, growth, cannibalism, and survival of juvenile crayfish. Soft tissue of adult female mussels was extracted with n-hexane, ethyl acetate, and methanol, and the methanolic extract (yield 5.5%) was analyzed by LC-HRMS. A compound annotated as ecdysterone was detected tentatively, together with diosgenin, drospirenone, and etiocholanolone. Juveniles (0.55 g) were reared for 40 days in a completely randomized design with four treatments (control; immersion at 1 mg L-¹ for 10 h every 5 days; diet with 2% extract; and both methods combined), three aquaria per treatment, and 30 animals per aquarium. The combined treatment gave the best results: molting frequency was 92% higher than in the control (2.30 vs 1.20 molts per individual), specific growth rate was 2.20% day-¹ (control 1.37% day-¹), cannibalism fell from 27.8% to 10.0%, and survival rose from 60.0% to 82.2%. Water quality was similar among treatments. Because a crude extract was used and the identity of ecdysterone was not confirmed with an authentic standard, these effects cannot be attributed to ecdysterone alone. The extract is a promising natural candidate for molting management of juvenile redclaw crayfish, and validation with purified ecdysterone, dose–response tests, and larger-scale trials are needed.
