Vitamin C Content in Daphnia sp. Enriched with L-ascorbic Acid Ethyl Cellulose at Different Enrichment Period
Abstract
Daphnia with a density of 10,000 ind./l medium were enriched with either 0.1, 0.5 or 1 g l-ascorbic acid ethyl cellulose (VC) per l culture medium. In this triplicate experiment, Daphnia were enriched with VC in a 12 litre aquaria for 0, 3, 6 or 9 h. Results showed that the maximum VC content in Daphnia enriched with 0.1, 0.5 and 1 g VC/l medium was 347, 471 and 482 mg VC/g Daphnia (dry basis), respectively. This VC content was obtained after 5 h, 5 h 12 min and 5 h 18 min of enrichment period. During 9 h enrichment period, the survival rate of Daphnia enriched with 0.1, 0.5 and 1 g VC/l medium was decrease linearly from 100% to 55.6%, 50.1% and 47.9%, respectively.

ABSTRAK
Dalam penelitian ini, Daphnia dengan padat tebar 10.000 ekor/l diperkaya dengan vitamin C (VC) pada tiga dosis yang berbeda, yaitu dosis 0,1, 0,5 dan 1 g VC/l media untuk masing-masing penelitian 1, 2 dan 3. Daphnia diperkaya dalam wadah volume 12 liter selama 0, 3, 6 dan 9 jam. Masing-masing perlakuan diulang sebanyak 3 kali ulangan. Hasil analisa respon menunjukkan bahwa waktu pengkayaan yang optimum untuk masing-masing dosis pengkayaan 0,1, 0,5 dan 1 g VC/l media secara berturut-turut adalah 5 jam, 5 jam 12 menit dan 5 jam 18 menit dengan kadar VC masing-masing sebesar 347, 471 dan 482 mg VC/g Daphnia. Tingkat kelangsungan hidup Daphnia sp. menunjukkan terjadinya penurunan secara linier dengan semakin bertambahnya waktu pengkayaan. Tingkat kelangsungan hidup Daphnia sp. setelah 9 jam masa pengkayaan ialah sebesar 55,63%, 50,12% dan 47,92% untuk dosis 0,1, 0,5 dan 1 g VC/l media.
