Abstract

A radioimmunoassay (RIA) for progesterone plasma of various animal species had been applied and validated in our laboratory. The method used antisera against 11 alpha hydroxy progesterone raised in goats (donated by Dr. L.E Edqvist, Sweden) in a 1:2000 dilution. A sensitivity of 10 pg/ml was obtained based on the readings for this concentration which was statistically different from a zeroconcentration (p<0.05). The intra-assay precision expressed as the coefficient of variation (CV) was 0.57%, 2.41% and 2.03% for the range of concentration 100-200, 200-300 and 300-400 pg/ml respectively. The between assay CV's measured from standard curves from different assays were 2.82%, 3.23%, 3.39%, 2.93%, 3.87%, 6.19%, 8.36%, 5.74% for the concentrations of 0, 5, 10, 20, 50, 100, 250 dan 500 pg/ml. A ecovery experinment was conducted to measure the accuracy of the system by using male plasma added to the sample containing known amounts of progesterone. A recovery of 86.56 +- 5.12% was obtained. Using this method, values for the normal cycling female swamp buffalo was obtained. They were 423 +- 60.3 pg/ml during the luteal phase and 84-253 pg/ml during the follicular phase. The concentration of progesterone obtained from Priangan ewes was 2.08 +- 0.5 pg/ml during the follicullar phase.
